Exosomes Functionally Deliver SiRNAs In Vivo To Repress Tumor Growth
SUZHOU, China , Sept. 5, 2023 /PRNewswire/ -- VesiCURE Therapeutics Co. Ltd., a biotechnology company based in Suzhou , China , recently released a BioRxiv preprint titled "Using Extracellular Vesicles for In Vitro SiRNA Delivery and in vitro". vivo" (doi: https://doi.org/10.1101/2023.08.26.554924). In this manuscript, the VesiCURE team presented their patented exosome-based technology called "dilEVry" and demonstrated its ability to deliver siRNA functional in a tumor model.
In recent years, exosomes have received considerable attention as a promising drug delivery vehicle in academic and biomedical settings. These native cell-derived nanoparticles have been successfully used to deliver antisense oligonucleotides (ASOs), proteins and small molecules in several clinical studies, demonstrating low immunogenicity and a good safety profile. However, its therapeutic potential for siRNA loading and delivery remains relatively unexplored.
Although the approval of different siRNA preparations is gaining momentum and the reach of RNAi is growing dynamically, delivery options for siRNA preparations remain limited. Achieving effective siRNA delivery in tumor tissues and non-hepatocyte cells still represents untapped potential worldwide.
In this study, the VesiCURE team used their platform 'dilEVry' to load tumor-suppressing siRNA exosomes. When administered endogenously to a CDX tumor model, this approach resulted in a gradual inhibition of tumor growth over time. Furthermore, a biodistribution study showed a significant accumulation of dye-labelled exosomes within tumors, which clearly indicates the potential of exosomes to deliver siRNA to tumors via intravenous infusion. This development is currently ongoing within the company.
The publication does not demonstrate systemic toxicity affecting body weight and provides sufficient evidence to support the ongoing development of electric delivery vehicles as a promising therapeutic option for various human diseases.
